Abstract
This study standardized a multiplex RT-qPCR protocol for detecting Influenza A virus in formalin-fixed, paraffinembedded (FFPE) tissues from suspected Severe Acute Respiratory Syndrome (SARS) cases. A total of 174 samples were analyzed with an adapted extraction method and a commercial respiratory virus assay. Influenza A was detected in 4.6% of cases, along with RSV, Influenza B, and SARS-CoV-2. The method proved feasible and robust for differential diagnosis in FFPE tissues, complementing immunohistochemistry. Multiplex RT-qPCR in post-mortem samples shows strong potential to improve etiological surveillance of fatal cases and support evidence-based public health decisions.
