Evaluation of commercial kits for detecting the antigen NS1-dengue – São Paulo

Abstract

 The dengue virus, which belongs to the family Flaviviridae, genus Flavivirus, consists of single-stranded RNA encoding structural and non-structural proteins. It has four serotypes: DENV-1, DENV-2, DENV-3 and DENV-4. The rapid laboratory diagnosis may greatly help to control the spread of the disease. The objective of the present work was to evaluate diagnostic kits for dengue, by detecting the NS1 protein, using virus isolation as reference. We used 147 sera samples of patients suspected of DENV infection, 64 were tested for virus isolation and 83 by capture ELISA IgM. Dengue NS1 Ag Strip (Bio- Rad) achieved a sensitivity of 89%, specificity of 66%, PPV of 67% and NPV of 88%. Dengue Duo Test (Cassette) showed a sensitivity of 89%, specificity of 68%, PPV of 70% and NPV of 88%. The Platelia Dengue NS1 ELISA Ag (Bio-Rad) had a sensitivity of 95%, specificity of 47%, PPV of 59% and NPV of 92%. Dengue Early ELISA kit (PANBIO) resulted in a sensitivity of 86%, specificity of 71%, PPV of 69% and NPV of 86%. In general, the evaluated kits can be used in diagnosis, associated with other criteria such as clinicalepidemiological or other laboratorial methods.
