Abstract
In this paper are presented the results of complement fixation test performed in 8455 sera obtained from unselected patients living in 59 communities in northeast of the State of São Paulo, Brazil. In this survey, 17,39% of the sera reacted with methanolic antigen of Trypanosoma cruzi. The referred antigen is a methanolic extract of T. cruzi heated at a temperature of 120o C., for 30 minutes. This antigen was found to keep its specific properties at room temperature and the isofixation curve with sera from proved cases of Chagas' disease is consistently of the type I; therefore, there is not a critical dose of antigen for complement fixation, permiting its use in a large and broad zone of dilutions. The T. cruzi were easily cultivated in a liquid and heated medium without red cells and protein precipitates. This medium gives good results in the cultivation of T. cruzi from infected animals.
