Abstract

Some granulomatous lesions in animals and in humans show the same histological pattern. However, there is no complete information on patho-biochemical alterations related to carbohydrate expression in Shistosoma mansoni-infected tissues. In the present study, it was shown that all tested lectins presented distinct staining patterns on both parasite egg-tegument and peri-ovular granuloma. The wheat germagglutin in (WGA) lectin showed an intense staining on egg-granuloma system (EGS) in the experimental schistosomiasis, whereas the WGA, Lotus tetragonolobus aggutinin (LTA) and peanut agglutinin (PNA) lectins stained only the S. mansoni egg in human patients material. The Ulex europaeus I agglutinin (UEAI) showed a weak and nonspecific staining pattern on EGS in both human and experimental models. On the other hand, the LTA lectin only stained the fibrosis ring of human hepatic granuloma. The intense staining pattern on both EGS and S. mansoni egg by WGA, and the solely S. mansoni egg staining by PNA lectin indicated the presence of n-acetil-glycosamine and galactose moieties, respectively. WGA, PNA, and Concanavalin agglutinin (Con A) failed to discriminate the cells type found in granuloma. In conclusion, lectin using-histochemical analysis is an important tool for investigating the specific biochemical alterations in experimental and human schistosomiasis.
