Abstract

This investigation aimed to evaluate the modified method performance of extraction of light filth from whole wheat flour, which was carried out at Food Microscopy Laboratory of Instituto Adolfo Lutz, São Paulo-SP by means of intra-laboratory study. For performing this study, the technique standardized and published by International AOAC, 2000 was modified by suppressing the reflux step for defatting the flour. The experimental design was as following: three spiked levels for insect fragments - FI (5, 15, and 30 FI per 50 g), and three for rodent hairs - RH (5,10, and 15 RH per 50 g) with six replicates for each level. Two analysts independently were performed the techniques. No change was observed in insect fragments recovery by means of the modified technique. Our results were of 97%, 96%, and 94% in average for FI, and of 78%, 77%, and 70% for RH in average. These results were higher when  compared with AOAC interlaboratorial study data for FI (94%, 86%, and 86%), but lower for RH recovery comparing to AOAC RH
data (95%, 91%, and 96%). A possible explanation for the low RH recovery in the present study might be on account of the use of magnetic stirring.
