Abstract

The aim of the present study was to standardize the use of blood collected on filter paper in seroepidemiological studies of human herpesvirus 8 (HHV-8) infection. HHV-8 latent and lytic antibodies were searched in eluates of blood collected on Schleicher Schuell membrane from 28 KS/AIDS patients and 10 health individuals without KS, using “in house” immunofluorescence assays (IFAs). Serum samples from the same individuals were previously evaluated by IFA–LANA and IFA-lytic, and the results were compared to the results obtained in this study. IFAs were conducted according to criteria established at Immunology Department of Instituto Adolfo Lutz, using BCBL-1 cells line stimulated or not with phorbol ester, and FITC-anti human IgG conjugate. Non-specific stainning on IFAs were detected when 1:50 and 1:100 blood eluated dilutions were employed. This unespecificity disappeared on subsequent blood dilutions. Concordant positive and negative results were obtained between sera and blood samples, but some minor differences were observed on antibodies title. The results obtained confirmed the use of blood collected on paper filter for HHV-8 seroepidemiological purposes.
